The process of bacterial conjugation is one of the methods of recombination. The conjugation in bacteria can be described as the transfer of DNA from one bacterial cell to another bacterial cell through cell to cell contact or by a bridge like connection between the two cells. This beneficial process was discovered for the first time by Joshua Lederberg and Edward Tatum in 1946. This process involves horizontal gene transfer like the transduction and transformation but these two processes do not involve direct cell-to-cell-contact. Bacterial conjugation is sometimes considered as sexual reproduction or mating as this process involves transfer of genetic material through cell contact. During this process the donor cell provides the mobilizable genetic element which is most probably a plasmid or trasnposon. Most conjugative plasmids ensure that the genetic material they are carrying is different from that present in the recipient cell. The genetic information that the recipient cell is going to receive is of great importance for it. The benefits may be in the form of antibiotic resistance, xenobiotic tolerance or the ability to use new metabolites. The plasmids carrying such information are called as bacterial endosymbionts.

The prototypical conjugative plasmid is the F-plasmid or F-factor. The F-plasmid is an episome with a length of 100 kb. Episome is a plasmid that can integrate itself into the bacterial chromosome through homologous recombination. This plasmid contains its own origin of replication known as oriV and an origin of transfer called as oriT. There can be only one copy of the f-plasmid in a given bacterium which may be either free or integrated and the bacterium with this copy is known as F-positive or F-plus. Cells lacking the F-plasmids are known as F-negative or F-minus. The F-negative cells function as recipient cells. The other genetic information that are loaded in the F-plasmids are tra and trb loci which are about 33 kb long and each carries about 40 genes. The tra locus carries a pilin gene and regulatory genes which are responsible for the formation of pili on the cell surface. The locus also contains genes for the proteins that attach themselves to the surface of the F-negative bacteria and initiate conjugation. There are some questions about the role of pili in conjugation as some scientists believe that the genetic transfer does not occurs through pili present on the cell surface. It has been shown that pili are denatured with SDS during conjugation. It has been shown by experiments that there are several proteins that code for tra and trb which open a channel between the bacteria and traD enzyme is thought to be responsible for this process. This enzyme is located at the base of pilus and it initiates membrane fusion.

After the initiation of conjugation by a signal an enzyme known as relaxase come into play. It creates a nick in one of the strands of conjugative plasmid at the oriT. This enzyme can work can act either alone or with the help of a dozen of proteins known as relaxosome. In the F-plasmid system the relaxase enzyme is known as TraI and the relaxosome is made up of TraI, TraY, TraM and the integrated host factor IHF. The nicked strand or the T-strand is then unwound from the broken strand and is transferred to the recipient cell in 5'terminus to 3'-terminus direction. The remaining strand may replicate either independent of conjugative action or in concert with conjugative action. Conjugative replication also requires a second nick before successful gene transfer may commence. The amount of the DNA transferred depends upon the duration of contact between the two bacterial cells. In common laboratory strains of E. coli the whole process completes in about 100 minutes. The transferred DNA can then be integrated into the recipient genome by homologous recombination.

Conjugation is a reliable method for transferring genetic material into a number of cells. In laboratory successful transfers have been carried out for bacteria to yeasts, plants, mammalian cells and isolated mammalian mitochondria. This process has advantages over other methods of genetic transfer.
